CommentsThe CD4+ T cell clone was isolated from a DR2-positive MS patient with a relapsing-remitting disease course. To stabilize the TCR-MHC/Epitope complex for crystallization; the epitope was covalently attached to the N-terminus of the TCR Vß domain via an octapeptide linker. There are four complexes in the crystallographic asymmetric unit. Because of their close similarity; the analysis of the TCR-MHC interactions applies to all four complexes. Clear electron density was observed for the interface between TCR and MHC/epitope in each of the four complexes. Several loops in the MHC ß2 chain and the TCR constant domains were disordered; but these regions are distant from the TCR-MHC interface. PDB chains for each complex are: Complex I : MHC ?-chain: A; MHC ß-chain: B; epitope chain: C; TCR ?-chain: M; TCR ß-chain: P. Complex II : MHC ?-chain: D; MHC ß-chain: E; epitope chain: F; TCR ?-chain: Q; TCR ß-chain: R. Complex III : MHC ?-chain: G; MHC ß-chain: H; epitope chain: I; TCR ?-chain: S; TCR ß-chain: T.
